If plants with variegated leaves are used, regenerated shoots will be of two types, green and albino. This oddity emphasizes that regenerants come from single cells and that the green and white sectors of the leaf have different plastid genotypes; the plant is a genetic chimaera. Of course albinos can only be maintained in culture, but sometimes they can be grown to quite a large size. Other plant material can be used for this experiment. The best of alternatives are probably tobacco (use leaf as explant) and cauliflower (use small florets as explant).

Page 9 Containment hoods, like laminar flow hoods, depend on air flow to maintain sterility in the work area. Because these hoods are designed to prevent the escape of cultured cells from the work area, however, the opening at the front of the hood is partially closed (generally by a window) and air travels from the top of the cabinet downward to intakes on the work surface. Air does not flow out of the hood into the lab environment without first being filtered to remove contaminants. The protection offered to workers by a containment hood is unnecessary for most plant tissue culture purposes, but the hood is very satisfactory for handling plant cultures.

Protoplast digestion mixtures generally include several enzymes, the concerted action of which efficiently releases cells from the cell wall. A mixture of cellulase, pectolyase, and macerase will be used in this experiment. Because naked plant cells are fragile, and can burst in solutions of low osmotic potential, mannitol is included in the digestion mixture as an osmoticum. Other ingredients included improve the stability of the protoplasts. The plant material used in the experiment is red onion.
